The protocol attached to the DevCells Kickoff Workshop DevNote does not include a description for how to prepare a cholesterol solution.
As far as I can tell, cholesterol is only available as a powder, what the best practice to resuspend it at the stated concentration of 50 mg/mL? And how does this differ from the PURE Cell protocol which does not include cholesterol?
Great question, thanks for pointing out the missing step of how to prepare the cholesterol stock solution - it’s sometimes too easy to take stock solutions for granted!
The addition of cholesterol confers some additional stability for the liposomes. Both protocols should yield good liposomes. Future updates to Nucleus will support spec sheets for multiple membrane modules
The cholesterol solution is prepared by just adding chloroform such that the stock concetration is 50 mg/mL. It should dissolve easily at this concentration.
